It was discovered in 1973 by Dr. Loren Pickart during research on human blood plasma, where it was observed that the albumin fraction from young donors stimulated liver cells from older donors to synthesize proteins characteristic of younger tissues.<\/p>\n<p>GHK-Cu occurs naturally in human plasma, saliva and urine. The concentration of this peptide in plasma changes with age &#8211; it is approximately 200 ng\/ml at the age of 20 and decreases to approximately 80 ng\/ml at the age of 60. This decrease correlates with the observed reduction in the body&#8217;s regenerative capacity, which makes GHK-Cu the subject of intensive research in the context of aging processes and tissue regeneration.<\/p>\n<h3>Mechanism of action<\/h3>\n<p>GHK-Cu is characterized by a unique mechanism of action that includes several interconnected molecular pathways:<\/p>\n<p><strong>Copper chelation<\/strong> \u2013 histidine in the peptide sequence forms a stable complex with the copper ion Cu\u00b2\u207a. Copper is a cofactor for key enzymes, including lysyl oxidase (LOX), which is responsible for the cross-linking of collagen and elastin, and superoxide dismutase (SOD), which has an antioxidant effect.<\/p>\n<p><strong>Gene expression modulation<\/strong> \u2013 research using Connectivity Map (Broad Institute) has shown that GHK-Cu affects the expression of thousands of human genes. This peptide activates genes related to DNA repair, extracellular matrix synthesis and the antioxidant response, while inhibiting pro-inflammatory genes.<\/p>\n<p><strong>Activation of signaling pathways<\/strong> \u2013 GHK-Cu interacts with integrin pathways (integrin \u03b11\u03b21), which affects cell adhesion, migration and proliferation. The peptide also stimulates the TGF-\u03b2 pathway involved in the healing and tissue remodeling processes.<\/p>\n<p><strong>Antioxidant effect<\/strong> \u2013 GHK has the ability to neutralize toxic products of lipid peroxidation, such as 4-hydroxynonenal (HNE) and acrolein, protecting cells against oxidative stress.<\/p>\n<h3>Directions of scientific research<\/h3>\n<p>GHK-Cu is the subject of extensive preclinical and clinical research. Major research areas include:<\/p>\n<p>Research on skin regeneration and wound healing<\/p>\n<p>Numerous in vitro and in vivo studies analyze the effect of GHK-Cu on wound healing processes. In animal models, accelerated wound contraction, increased angiogenesis and stimulation of collagen synthesis were observed. Studies on human fibroblasts have shown increased production of type I and III collagen, elastin and glycosaminoglycans. Hydrogel and nanoparticle formulations containing GHK-Cu are of particular interest and are being tested for application in new generation dressings.<\/p>\n<p>Dermatological and cosmetic tests<\/p>\n<p>Clinical studies involving women aged around 50 showed that the application of GHK-Cu creams for 12 weeks influenced the density and thickness of the skin, reduced fine wrinkles and improved elasticity. Comparative clinical studies analyzed the effectiveness of GHK-Cu in relation to vitamin C and retinoic acid in the context of stimulating collagen production.<\/p>\n<p>Pulmonary fibrosis research<\/p>\n<p>In mouse models of bleomycin-induced pulmonary fibrosis, GHK showed the ability to reduce the infiltration of inflammatory cells, reduce the thickness of interstitial tissue and reduce the level of pro-inflammatory cytokines (TNF-\u03b1, IL-6). These studies suggest potential applications in the context of idiopathic pulmonary fibrosis through modulation of the TGF-\u03b21\/Smad pathway.<\/p>\n<p>Research on the function of fibroblasts<\/p>\n<p>Research on the effect of GHK-Cu on fibroblasts from patients with chronic obstructive pulmonary disease (COPD) is particularly interesting. GHK treatment restored the ability of these cells to contract and remodel the collagen gel, suggesting potential in restoring the regenerative functions of damaged tissues.<\/p>\n<p>Hair growth research<\/p>\n<p>Preclinical studies analyze the effects of GHK-Cu on hair follicles. This peptide stimulates the proliferation of hair papilla cells and extends the anagen (growth) phase of the hair cycle. Work is underway on formulations for local application in the context of androgenic alopecia.<\/p>\n<p>Neuroprotective studies<\/p>\n<p>Gene expression profile analyzes indicate the effect of GHK on genes related to neuronal function and DNA repair. In the context of neurodegenerative diseases, the potential of GHK-Cu is being investigated as a regulator of copper homeostasis, the disturbance of which is observed in Alzheimer&#8217;s disease.<\/p>\n<h3>Scientific context<\/h3>\n<p>GHK-Cu is part of a growing class of bioactive peptides being studied in regenerative medicine. It stands out from other cosmetic peptides (such as Matrixyl or Argireline) due to its endogenous origin and broad biological activity profile confirmed by molecular tests.<\/p>\n<p>The stability of GHK-Cu is both an advantage (high stability in solutions) and a research challenge &#8211; the peptide is susceptible to degradation by carboxypeptidases present in wound fluids. For this reason, research is being conducted on advanced delivery systems, including nanoparticles, hydrogels and conjugates with cell-penetrating peptides.<\/p>\n<h3>Research safety profile<\/h3>\n<p>In available preclinical and clinical studies, GHK-Cu is characterized by a favorable safety profile. No significant side effects were noted in dermatological studies with topical application. The peptide is a natural component of the human body, which theoretically reduces the risk of immune reactions.<\/p>\n<p>However, it should be noted that excessive copper concentrations may lead to unfavorable effects, including increased activity of matrix metalloproteinases (MMP), which in excess may contribute to collagen degradation.
