What is tirzepatide?

Tirzepatide (LY3298176) is an innovative research peptide developed by Eli Lilly, representing a new class of incretin compounds. It is the first ever dual agonist of GIP and GLP-1 receptors (so-called "twincretin"), which simultaneously activates two key metabolic pathways: the glucose-dependent insulinotropic peptide (GIP) receptor and the glucagon-like peptide 1 (GLP-1) receptor.

This synthetic peptide consists of 39 amino acids and was designed based on the GIP skeleton with the addition of structural modifications. Due to acylation with the albumin-binding C20 fatty acid moiety, tirzepatide has an extended half-life of approximately 5 days, enabling once-weekly administration in clinical trials.
